FWIW, playing guitar then going to Rock Band/Guitar Hero is totally different. I'm far to used to moving up and down a fretboard, so playing to 5 keys is a bit of a change.
Conversely, my cousin, who plays nothing more than the radio continually kicks my ass at Guitar Hero. But he couldn't play a guitar with a gun to his head.

I've always wanted to see a band play its song on Rock Band... And +1 Turd, if anything -not- being able to play a real instrument is an asset, because you don't get those "WTF" moments when you try to do real-instrument stuff with a game controller.

I play the drums both in RL and in Rock Band. It gets tough sometimes, especially when I know the song that is being played. I think to myself, "I can really play this song but faking it is kicking my ass!"
